What is the probability of picking 2 vowels from this set of 6 tiles?

Probability is the count of outcomes we desire, divided by the count of total outcomes. So that's the number of ways to pick 2 vowels out of 3, divided by the number of ways to pick any 2 tiles.

2 vowels out of 3 is expressed as 3 choose 2. 2 tiles out of 6 is expressed as 6 choose 2. So our probability will be 3 choose 2, divided by 6 choose 2

Which is 3 over 15, or 1 over 5.
